The active form of Vitamin D3, Calcitriol, works by binding to Vitamin D receptors (VDRs), which are present in various body tissues. Vitamin D3 has a long half-life of approximately 50 days due to its fat-soluble nature. It is absorbed in the small intestine, where it binds to specific α-globulins and is transported to the liver. In the liver, it undergoes metabolism to 25-hydroxy Vitamin D3 (Calcidiol). Further hydroxylation occurs in the kidneys, converting it into 1,25-dihydroxy Vitamin D3 (Calcitriol), the metabolite that enhances calcium absorption. Any unprocessed Vitamin D3 is stored in fat and muscle tissues. Vitamin D3 is primarily excreted through feces and urine.
